Here's what I do for file sizes.

I use iMovie to import my videos and materials and save them using 
the following settings:

Image Settings = 240x180,
Compression = Motion JPEG A
Key Frame Rate = 0
Audio Compression = IMA 4:1
Sample Rate 22050 per second
Sample Size = 16 bits
Channels = Stereo

A one hour video takes approximately 17 GB with these settings. I 
then go to Terran's (now Discreet or Media100's) Cleaner 5.1.1 and 
apply the following settings:

Output = QuickTime (.mov), Flatten/cross-platform/fast-start, 
Compress movie headers, Prepare for Streaming
Tracks = Process video, Process audio, Process MPEG
Image = Manual crop, Image size 400x300, Accurate scaling, 
Deinterlace (auto), Noise Reduce
Adjust  = Brightness 15, Black Restore 20
Encode = Sorenson, Millions of Colorrs, Spatial 50%, 12 fps, Keyframe every 24
Sorenson = Highest Quality (normal speed), Smoothing
Audio = QDesign Music 2, 3.0kBytes/s, 16-bit mono, samples at 
22.050KHz, adjust volume to 125%, noise removal
Alternate = T1/cable modem, quality 8, computer power 400, QuickTime 4
Metadata = Software: Encoded with Cleaner 5

These settings have given me the ability to compress that one hour 
video down to about 535MB with a tolerable loss of quality for the 
length and size of the final product.
